Challenges in Peptide Synthesis
Nowadays, well-known high-yielding protocols are available for the (automated) coupling of amino acids and the protection/deprotection steps. However, it is still not always straightforward to make specific high purity peptides, often because of following challenges:
- Synthesis of long peptides (> 30 amino acids), especially highly hydrophobic
- Synthesis on large scale
- Influence of coupling agent and side chain on racemization
- Determination of (optical) purity
